چکیده

The process of ultrasonic-enhanced compound-enzymes-assisted extraction of polysaccharides from the fruit of Cornus officinalis was optimized by response surface methodology (RSM). The influence of three different factors on the yield of C. officinalis polysaccharides (COP) was studied. The results showed that the optimal conditions were extraction temperature 49.6 °C, ultrasound time 40.41 min, and ultrasound power 308.07 W. The experimental data obtained were fitted to a second-order polynomial equation using multiple regression analysis and were analyzed by analysis of variance (ANOVA). The 3-D response surface plot and the contour plot derived from the mathematical models were applied to determine the optimal conditions. Under modified conditions, the experimental yield of polysaccharides was 11.02±0.41%, which was well in agreement with the value predicted by the model. A good 1,1-diphenyl-2-picryldydrazyl (DPPH) radical scavenging activity of COP extracted by ultrasonic-enhanced compound-enzymes-assisted extraction was observed.

A Process Development FOR Extraction of Marker Compound from EMBLICA OFFICINALIS FRUIT

The present study is done to examine and optimize the process of enzyme-assisted extraction of Gallic acid from the dried fruit of Emblica officinalis. The aqueous extraction yielded 12.09% of Gallic acid. A selective extraction process after the treatment with enzymes is proposed by using 30% (v/v) methanol which releases up to 15.68% of the Gallic acid, present in the dried fruit. The optimal...

Hepatoprotective and Antioxidative Activities of Cornus officinalis against Acetaminophen-Induced Hepatotoxicity in Mice

The fruit of Cornus officinalis Sieb. et Zucc. is commonly prescribed in Asian countries as a tonic formula. In this study, the hepatoprotective effect of ethanolic extracts of the fruit of C. officinalis (ECO) was investigated in a mouse model of acetaminophen- (APAP-) induced liver injury. Pretreatment of mice with ECO (100, 250, and 500 mg/kg for 7 days) significantly prevented the APAP (200...

A SCAR marker for the analysis of chloroplast DNA from different cultivars of Cornus officinalis.

The aims of this study were to establish a random amplified polymorphic DNA (RAPD) fingerprint database of chloroplast DNA (cpDNA) from different cultivars of Cornus officinalis and to convert RAPD markers to sequence characterized amplified regions (SCAR) markers. A method of extraction was established that was suitable for obtaining cpDNA from samples rapidly dried in silicone; an RAPD finger...

Effect of Cornus Officinalis on Receptor Activator of Nuclear Factor-kappaB Ligand (RANKL)-induced Osteoclast Differentiation

OBJECTIVES Osteoporosis is a disease of bones that is thought to result from an imbalance between bone resorption and bone formation. Although osteoporosis itself has no symptoms, osteoporosis caused by osteoclasts leads to an increased risk of fracture. Here we examined the effects of cornus officinalis on receptor activator of nuclear factor-kappaB ligand (RANKL)-mediated osteoclast different...
